Cellules souches de la pulpe dentaire et matrice de collagène : une association au service de l’ingénierie tissulaire osseuse prévascularisée

Abstract : Critical-size defects overcome the natural ability of the bone tissue to heal. Currently, all the therapeutics including autologous grafting are facing limitations. The answer could be provided by bone tissue engineering with the creation of a prevascularized bone substitute that could allow both revascularization of the graft and new bone formation. Our project aimed to develop this prevascularized bone substitute by combining dental pulp stem cells (DPSCs), and collagen matrix enriched with alginate and HA hydrogel. First, we highlighted the preferred experimental condition for the use of DPSCs in bone tissue engineering. We showed that DPSCs are not affected by lecithin rapeseed nanoliposomes (which can be used for the vectorization of bioactive molecules), and that a low glucose concentration can induce osteogenic differentiation as effectively as a high glucose concentration. Then, we seeded DPSCs in biomaterials composed of collagen matrix with alginate and HA hydrogels and obtained both osteogenic differentiation and endothelial-like cells in our constructs. We also showed that alginate/HA hydrogel had a booster effect on the cellular colonization and differentiation in our biomaterials compared to collagen alone. These encouraging results highlight the potential of DPSCs combined with a collagen and alginate-HA hydrogel composite construct in prevascularized bone tissue engineering.
